Tiffany Thompson will be presenting her essay on the 9th floor of the Center for Vaccine Research during one of her collaborative lab meetings. Her essay, A Literature Review of the Histology of Rift Valley Fever Virus Neurological Disease Comparing Rodent and NHP Models to Human Disease, will focus on the neurological disease seen in the developed animal models and the histology being used in the research of the Rift Valley Fever Virus.

Dana Woell's thesis, "Impact of H5N1 Influenza Virus Infection of Macaques on Natural Killer Cells and Innate Lymphoid Cell Populations" has demonstrated in a nonhuman primate model that aerosolized infection with H5N1 influenza virus leads to disease progression similar to that seen in human cases.  This study aims to characterize some of the innate immune cells that contribute to the response to severe H5N1 infection in this macaque model.

Pranali Ravikumar's thesis, "Programming Dendritic cells for Intercellular Delivery of T-bet to Enhance Function of Cytotoxic T Cells" will show that the immune mechanism of DC1 to CTL intercellular transfer can be exploited to enhance anti-HIV T cell response, or to correct their dysfunction of T cell exhaustion as supported by evidence in DC1-based cancer immunotherapy studies.

Joe Albe's thesis "Viral Encephalitis: Phenotyping Leukocyte Infiltration into the Central Nervous System as a Result of Rift Valley Fever Virus Infection"  will show you how the findings of aerosol infected rats mainly exhibit innate leukocyte infiltration to the brain while subcutaneous have limited leukocyte infiltration and display a stronger adaptive immune response. The relative contributions of viral cytopathology versus immunopathology remain to be determined, but this study represent the first attempt to characterize the leukocytic component of RVFV infection.

A supervised consumption site is a legally sanctioned health facility that offers a hygienic environment where people can use illicit drugs under the supervision of trained staff.  Although over 90 facilities have been implemented internationally to date, only two have been established in North America, both in Vancouver.  There is renewed interest across Canada and the US in supervised injection given the current marked rise in opioid overdoses and related deaths.  This talk will include discussion of how the intersection of evidence, research and policy moved the discussion of supervised injection in Toronto from a controversial idea to a widely supported policy recommendation with anticipated approval within the next few weeks.  This talk will focus on how modeling can inform decisions about uncertain and controversial interventions and thoughts about conducting policy-relevant research that is controversial and value-laden.

Henry Ma's thesis "Investigation of the Natural History of Equine Encephalitis Viruses with Radiofrequency Telemetry for Detection of Subclinical Disease Patterns" seeks to establish and study a model of human equine encephalitis virus infection via the aerosol route in the nonhuman primate, the cynomolgus macaque (Macaca fascicularis) with a focus on the natural history of disease through examination of radiofrequency electrocardiography data, and evaluates the feasibility of the use of such data to prognosticate severe disease courses which include encephalitis.

Network analysis supports the assessment of power and influence within and between groups, and identification of key actors and ideas.  In dynamic network analysis, this is extended to look at changes over time and space in these networks.  In this talk, the nature of dynamic network analysis is described as are the challenges to this form of analysis posed by massive data.  A set of application problems from the health area or relevant to health data is used for illustration.

INHALATIONAL MODELS OF RIFT VALLEY FEVER: KEYS TO UNDERSTANDING FATAL VIRAL ENCEPHALITIS

Amy Hartman, PhD, Assistant Professor of Infectious Diseases and Microbiology

Hartman’s broad research interests center on understanding the pathogenic mechanisms of highly virulent RNA viruses, particularly those that present an emerging threat to global health. A major focus of her research program is on Rift Valley Fever virus (RVFV), a virus that causes severe disease in livestock and humans in Africa and the Arabian Peninsula. RVFV is a mosquito-borne virus that has the potential to emerge in developed countries and cause significant damage to the livestock industry, as well as human morbidity and mortality. RVFV is also a potential biological warfare agent due to its infectiousness through inhalation. Encephalitis caused by RVFV has been vastly understudied in the past due to the lack of suitable animal models. With funding from the Department of Defense (DoD), Hartman’s lab characterized the first rat and monkey models of neurological disease seen in some RVFV-infected humans. These animal models are the tools that are needed to understand the neuropathogenesis of this disease.

There are important gaps in our understanding of the global burden and spatial transmission dynamics of influenza. Based on retrospective analysis of a variety of epidemic and pandemic seasons in the Americas, Europe, Africa and Asia. Dr. Viboud will discuss how disease burden may vary with geography due to differences in seasonality, underlying socio-economic conditions, and co-circulation of other respiratory and non-respiratory pathogens. She will present recent modeling work on the spatial transmission of epidemic and pandemic viruses and the association with demographic factors and human mobility. There are important gaps in our understanding of the global burden and spatial transmission dynamics of influenza. Based on retrospective analysis of a variety of epidemic and pandemic seasons in the Americas, Europe, Africa and Asia. Dr. Viboud will discuss how disease burden may vary with geography due to differences in seasonality, underlying socio-economic conditions, and co-circulation of other respiratory and non-respiratory pathogens. She will present recent modeling work on the spatial transmission of epidemic and pandemic viruses and the association with demographic factors and human mobility. If time permits, she will highlight key findings of a recent infectious disease forecasting challenge led by Fogarty/NIH in the context of the Ebola response, which has just concluded.

The value of simple models, rapidly built with available data, to aid decision makers during response to catastrophic infectious disease events will be demonstrated through 4 models: 1) Estimating, in near real-time, the impact of the 2009 H1N1 influenza pandemic; 2) Modeling input to aid the decision regarding the production of H3N2v monovalent vaccine; 3) Estimating impact of proposed vaccination for a hypothetical H5N2 influenza pandemic (exercise); and 4) A Risk-Benefit analysis of different options for testing and treating asymptomatic patients potentially infected with contaminated steroid product. These examples will be used to illustrate some basic concepts that must guide modelers as they interact with public health decision makers during responses.

Interactions arising from sequential viral and bacterial infections may play important roles in the epidemiological outcome of many respiratory pathogens. Influenza virus has been implicated in the pathogenesis of several respiratory bacterial pathogens commonly associated with pneumonia. However, studies focusing on different scales of resolution have painted an inconsistent picture. Individual-scale animal experiments have unequivocally demonstrated an association, whereas the support in population-level epidemiological studies remains variable and unclear. We use multiple sources of epidemiological data which vary in the time periods they cover and the scales at which they are collected, and a mechanistic transmission model, both within a likelihood-based inference framework to characterize the nature, timing, and magnitude of this interaction. We find support for a strong but short-lived interaction, with influenza infection increasing susceptibility to pneumococcal (and other bacterial) pneumonia by ~100-fold, in line with our understanding from animal challenge experiments. The population-level impact of such an interaction can be modest during non-pandemic seasonal influenza periods (which may in turn make it difficult to detect), but can be substantially larger during pandemic periods (and also more detectable). We argue that these seemingly inconsistent observations at different scales can be explained by this proposed interaction: a strong but short-lived susceptibility enhancement of pneumonia following influenza.
